Expression and Promoter Analysis of Six Heat Stress-Inducible Genes in Rice
نویسندگان
چکیده
During the long evolutionary process, plant gradually formed a series of strategies and mechanisms to cope with stress environment such as drought, heat, cold, and high salinity. Six highly heat responsive genes were identified in rice by microarray data analysis. The qRT-PCR analysis confirmed that the expression of these six genes were highly heat inducible and moderately responded to salt stress, polyethylene glycol, and abscisic acid treatment, but little affected by cold treatment. Promoters of the three highly heat-inducible genes (OsHsfB2cp, PM19p, and Hsp90p) were used to drive GUS gene expression in rice. The results of the GUS gene expression, histochemical staining, and GUS activities in panicles and flag leaves of the transgenic rice plants confirmed high heat-induced GUS activities and moderate drought-induced activities. The three promoters exhibited similar high activity lever in rice leaf under heat, but OsHsfB2cp and PM19p showed much higher activities in panicles under heat stress. Our work confirmed that the OsHsfB2c and PM19 promoters were highly heat inducible and further characterization and reconstruction of cis-elements in their promoters could lead to the development of highly effective heat-inducible promoters for plant genetic engineering.

Evaluation of MYB93 and MAD8 Genes in Transgenic and Non-Transgenic Rice
Increasing drought tolerance, especially in rice, which is one of the most important crops in Asia, is necessary. Transcription factors are specific sequence DNA-binding proteins that are capable of activating or suppressing transcription. These proteins regulate gene expression levels by binding to cis regulatory elements in the promoter of target genes to control various biological processes ...
متن کاملMonitoring expression profiles of rice genes under cold, drought, and high-salinity stresses and abscisic acid application using cDNA microarray and RNA gel-blot analyses.
To identify cold-, drought-, high-salinity-, and/or abscisic acid (ABA)-inducible genes in rice (Oryza sativa), we prepared a rice cDNA microarray including about 1700 independent cDNAs derived from cDNA libraries prepared from drought-, cold-, and high-salinity-treated rice plants. We confirmed stress-inducible expression of the candidate genes selected by microarray analysis using RNA gel-blo...

Sclerotinia sclerotiorum is a phytopathogenic fungus which causes serious yield losses in canola. A pathogen inducible-promoter can facilitate the production of Sclerotinia-resistant transgenic canola plants. Inthis study, the “gain of function approach” was adopted for the construction of a pathogen-inducible promoter. The synthetic promoter technique was used, which involved the in...
متن کاملThe Arabidopsis transcriptional regulator DPB3‐1 enhances heat stress tolerance without growth retardation in rice
The enhancement of heat stress tolerance in crops is an important challenge for food security to facilitate adaptation to global warming. In Arabidopsis thaliana, the transcriptional regulator DNA polymerase II subunit B3-1 (DPB3-1)/nuclear factor Y subunit C10 (NF-YC10) has been reported as a positive regulator of Dehydration-responsive element binding protein 2A (DREB2A), and the overexpressi...

water stress and, in this regard, it is necessary to improve rice cultivars to tolerance to environmental stresses. In this research 154 recombinant inbred lines (F9) derived from a cross between Shah-Pasand and IR28 in three conditions (non-stress, osmotic stress -0.3 and -0.6 Mpa induced through polyethylene glycol-6000) were evaluated as a factorial experiment in randomized complete block de...
